Could Fish Oil Potentially Cure Leukemia?

The results of a new study just published in the Journal Blood, has highlighted how a compound found in fish oil may in future be able to eradicate the blood cancer known as leukemia.
In laboratory experiments at Pennsylvania State University, researchers found that a component of Omega 3 known as Delta-12-J3 Prostaglandin or D12PGJ3 can selectively target the stem cells of chronic myelogenous leukemia or CML.
D12PGJ3 arises from Eicosapentaenoic acid (EPA), long chain fatty acid found abundantly in oily fish. It has now been shown in plenty of prior studies that Omega 3 fatty acids have important health benefits in helping to lower the risk of disease but this latest trial now opens up the incredible possibility that Omega 3 may be used in cancer therapy at some point.
"Research in the past on fatty acids has shown the health benefits of fatty acids on cardiovascular system and brain development, particularly in infants but we have shown that some metabolites of Omega-3 have the ability to selectively kill the leukemia-causing stem cells" said Sandeep Prabhu, author of the new study and associate professor of immunology and molecular toxicology in the Department of Veterinary and Medical Sciences
Prabhu also said that it's necessary to target the stem cells as they can divide and cause more cancer cells.
Current drugs used for CML can prolong a patient's life by keeping the number of leukemia cells in the blood down but they can't cure leukemia because they don't specifically target the leukemia stem cells explained Robert Paulson, Co author of the study and associate professor of veterinary and biomedical sciences.
"The patients must take the drugs continuously," said Paulson. "If they stop, the disease relapses because the leukemia stem cells are resistant to the drugs."
"These stem cells can hide from the treatment, and a small population of stem cells give rise to more leukemia cells," said Paulson.
"So, targeting the stem cells is essential if you want to cure leukemia."
CML is a relatively rare kind of cancer that results in too many white blood cells being produced. It is not a disease passed down through families and can occur in any individual at any age although it's more likely to affect middle aged and older people.
The Leukemia begins in the bone marrow in a cell called a stem cell. Stem cells produce new blood cells by making a copy of themselves and then dividing to make two new cells. In most cases of this disease, something goes drastically wrong during this process of cell division and genes that are normally in separate chromosomes get stuck together producing an abnormal cell. This gene mutation is called the Philadelphia chromosome. However, the existence of the Philadelphia Chromosome is not the only factor used to make a diagnosis of CML as it is often present in other types of leukemia.
Tests performed to diagnose CML include a full blood count and platelet count, a bone marrow biopsy, identification of the Philadelphia chromosome, and physical examination to detect if the spleen is swollen.
Exposure to radiation can increase the risk of developing CML although in most incidences of leukemia the precise cause is not apparent.
There are three phases of CML. The first phase is called the chronic phase and this stage develops slowly over many years. During this period there may be no symptoms. The second phase is the accelerated phase and during this stage there may be pain in the left side under the ribcage, mild fever and bone pain. The end phase is called the blast crisis and this is the terminal stage.
If the disease is caught soon enough then there are drugs available which can stop the progress of the disease but the only cure is a bone marrow transplant. Many people are diagnosed during the last stage. During this phase there is nothing that can be done.
The scientists in this latest study are now working to find out if the compound found in fish oil can be used to treat the blast crisis stage of this kind of Leukaemia. They have also applied for a patent and intend to test the fish oil compound in human studies next.

NEW YORK (Reuters Health) - Older adults who took vitamin B12 and folic acid supplements for two years had greater improvements on short- and long-term memory tests than adults who did not take the vitamins, according to the results of a new study from Australia.
The benefits were modest, but encouraging, indicating that the vitamins "may have an important role in promoting healthy ageing and mental wellbeing, as well as sustaining good cognitive functioning for longer on a community-wide scale," Janine Walker, the lead author of the study and a researcher at Australian National University, told Reuters Health by email.
The researchers asked more than 700 people, aged 60 to 74 years, to take a daily dose of folic acid and vitamin B12 or fake pills that resembled the vitamins. The study only included people who showed signs of depression, but were not diagnosed with clinical depression.
The vitamin dose included 400 micrograms of folic acid and 100 micrograms of vitamin B12. The participants didn't know which pills they were assigned to take.
"We felt that older people with elevated depressive symptoms were an important cohort to target given evidence that late-life depression is associated with increased risk of cognitive impairment," Walker said.
After 12 months, there seemed to be no difference between the groups in how well the people scored on mental tests, including memory, attention and speed.
Two years on, however, those who took the vitamins showed larger improvements in their scores on the memory tasks.
The difference in the improvements was small, the researchers write in their study, published in the American Journal of Clinical Nutrition.
For instance, on a short term memory test, those who took the fake pills improved their score from about 5.2 to about 5.5 over the two years. Those who took the vitamins increased their test scores from 5.16 to roughly 5.6.
Short term memory is used to dial a number someone has just told you, while long term memory comes into play when you try to call that number a day or week later.
Joshua Miller, a professor at the University of California, Davis, said it's difficult to translate the memory improvements on the tests into real life benefits. He said it's likely that some people had larger memory improvements, while others benefited very little.
"For any given individual, there may or may not be an effect. But on a population level, a small increase in cognitive function can have very real ramifications on the functioning of the population as a whole and on the costs of healthcare." Miller said.
Miller said it's possible that certain subgroups of individuals might be more likely to benefit from folic acid and B12 than others.
"What I believe the next thing we need to do is (study) the group that is most likely to benefit from this," Miller, who did not participate in this research, told Reuters Health.
UNCLEAR HOW VITAMINS MIGHT WORK
Walker said it's not clear yet how adding vitamins might work to boost brain functioning, and not all studies have agreed upon their benefits.
One idea is that the vitamins reduce the body's levels of a molecule called homocysteine, which is linked to cardiovascular disease and poor cognitive function.
The body uses homocysteine to build proteins, but high levels of it in the blood are associated with heart disease -- and heart disease is linked to mental decline.
The thinking goes that lowering homocysteine could perhaps reduce someone's cardiovascular risk, and in turn affect his mental functioning.
A recent test of folic acid's influence on cardiovascular disease, however, found that in people with high homocysteine levels, the vitamin didn't help prevent thickening of the arteries (see Reuters Health report of April 13, 2011).
Walker said it will be important to test whether other groups of people, especially those older than the people who participated in this study, could also benefit from taking the supplements.

'Supercentenarians' appear to have built-in protection against disease, study finds

SUNDAY, Jan. 8 (HealthDay News) -- People who live 110 years or longer have as many disease-associated genes as those in the general population, but they may also be blessed with protective genes that help them live so long, researchers report.
The team of U.S. scientists noted that supercentenarians, as they are called, are extremely rare, with only one per 5 million people in developed nations. There is growing evidence that genetics play a major role in living to such an old age.
In what they describe as a first-of-a-kind study, the researchers analyzed the whole genome sequences of a man and a woman who lived past the age of 114 and found that they had as many disease-associated genes as other people.
For example, the man had 37 genetic mutations associated with increased risk for colon cancer.
"In fact, he had presented with an obstructing colon cancer earlier in his life that had not metastasized and was cured with surgery. He was in phenomenal cognitive and physical shape near the time of his death," study senior author Dr. Thomas Perls, director of the New England Centenarian Study, said in a Boston University Medical Center news release.
The woman had numerous genetic variations associated with age-related disease, such as heart disease, cancer and Alzheimer's disease. She did develop congestive heart failure and mild cognitive impairment, but these conditions didn't become evident until she was more than 108 years old.
"The presence of these disease-associated variants is consistent with our and other researchers' findings that centenarians carry as many disease-associated genes as the general population," Perls said. "The difference may be that the centenarians likely have longevity-associated variants that cancel out the disease genes. That effect may extend to the point that the diseases don't occur -- or, if they do, are much less pathogenic or markedly delayed towards the end of life, in these individuals who are practically living to the limit of the human lifespan."
The study was published Jan. 3 in the journal Frontiers in Genetics, and researchers will be able to access the information at the U.S. National Institutes of Health data repository.

Leukoencephalopathy with brainstem and spinal cord involvement and lactate elevation (commonly referred to as LBSL) is a progressive disorder that affects the brain and spinal cord. Leukoencephalopathy refers to abnormalities in the white matter of the brain, which is tissue containing nerve cell fibers (axons) that transmit nerve impulses.
Most affected individuals begin to develop movement problems during childhood or adolescence. However, in some individuals, these problems do not develop until adulthood. People with LBSL have abnormal muscle stiffness (spasticity) and difficulty with coordinating movements (ataxia). In addition, affected individuals lose the ability to sense the position of their limbs or vibrations with their limbs. These movement and sensation problems affect the legs more than the arms, making walking difficult. Most affected individuals eventually require wheelchair assistance, sometimes as early as their teens, although the age varies.
People with LBSL can have other signs and symptoms of the condition. Some affected individuals develop recurrent seizures (epilepsy), speech difficulties (dysarthria), learning problems, or mild deterioration of mental functioning. Some people with this disorder are particularly vulnerable to severe complications following minor head trauma, which may trigger a loss of consciousness, other reversible neurological problems, or fever.
Distinct changes in the brains of people with LBSL can be seen using magnetic resonance imaging (MRI). These characteristic abnormalities typically involve particular parts of the white matter of the brain and specific regions (called tracts) within the brain stem and spinal cord, especially the pyramidal tract and the dorsal column. In addition, most affected individuals have a high level of a substance called lactate in the white matter of the brain, which is identified using another test called magnetic resonance spectroscopy (MRS).

How common is LBSL?

LBSL is a rare condition. Its exact prevalence is not known.

What genes are related to LBSL?

LBSL is caused by mutations in the DARS2 gene, which provides instructions for making an enzyme called mitochondrial aspartyl-tRNA synthetase. This enzyme is important in the production (synthesis) of proteins in cellular structures called mitochondria, the energy-producing centers in cells. While most protein synthesis occurs in the fluid surrounding the nucleus (cytoplasm), some proteins are synthesized in the mitochondria.
During protein synthesis, in either the mitochondria or the cytoplasm, building blocks (amino acids) are connected together in a specific order, creating a chain of amino acids that forms the protein. Mitochondrial aspartyl-tRNA synthetase plays a role in adding the amino acid aspartic acid at the proper place in mitochondrial proteins.
Mutations in the DARS2 gene result in decreased mitochondrial aspartyl-tRNA synthetase enzyme activity, which hinders the addition of aspartic acid to mitochondrial proteins. It is unclear how the gene mutations lead to the signs and symptoms of LBSL. Researchers do not understand why reduced activity of mitochondrial aspartyl-tRNA synthetase specifically affects certain parts of the brain and spinal cord.

How do people inherit LBSL?

LBSL is inherited in an autosomal recessive pattern, which means both copies of the gene in each cell have mutations. In this condition, each copy of the gene carries a different mutation (compound heterozygous mutations). An affected individual never has the same mutation in both copies of the gene (a homozygous mutation). The parents of an individual with an autosomal recessive condition each carry one copy of the mutated gene, but they typically do not show signs and symptoms of the condition.
